Inhibition of radiation-induced changes of glyoxalase I activity in mouse spleen and liver by phenothiazines.

Swiss albino mice (male) were irradiated with gamma-rays at a dose-rate of 0.05 Gy s-1, and the activities of glyoxalase I (GI) and glyoxalase II (GII) were determined after 24 h in the spleen and liver. Radiation up to 4 Gy increased the activity of GI and decreased that of GII. It was possible that the radiation-induced changes in the activity of the glyoxalase system, particularly that of GI, were suggestive of the regeneration status of the tissue. Phenothiazines such as chlorpromazine (CPZ), promethazine (PMZ) and trimeprazine (TMZ) inhibited the radiation-enhanced activity of GI in a concentration-dependent manner. On the other hand, almost no change in the activity of GII was observed using phenothiazines. The effect of phenothiazines on radiation-induced changes of glyoxalase activity were reversed in the presence of ferrous (Fe2+) ions. However, phenothiazines inhibited the radiation effect in the presence of ferric (Fe3+) ions. This combined effect was predominant in the liver. A possible mechanism for the modifying effect of phenothiazines is suggested.

Safety, efficacy, and acceptance of intramuscular sedation: assessment of 900 dental cases.

Intramuscular (IM) sedation techniques remain the least used by clinicians, possibly because of their limited exposure in the undergraduate and graduate dental curricula. To assess the safety and efficacy of these techniques, the authors conducted a retrospective study of 900 dental cases. A total of 308 patients received ketamine, 356 received a meperidine/promethazine combination, and 236 received midazolam. Results revealed a high safety record, efficacy, and good evaluations from clinicians and patients, in addition to some advantages over other sedation techniques.

[Advances in the pharmacotherapy of osteoporosis]

In the pharmacotherapy of osteoporosis administration of calcium, small amounts of vitamin D and fluorides still is the basic treatment. Thyrocalcitonin can be also administered, recently also in the form of a nasal spray. It does not only prevent reabsorption of bone but has also an analgetic effect. In the treatment of postmenopausal osteoporosis we use conjugated oestrogens in cyclic preparations with gestagens. Biphosphonates prevent bone resorption but also apposition of the mineral. As to new preparations, Ipriflavone and TGF-beta seem most promising, moreover, there is the perspective of the use of other preparations: small doses of PTH, tamoxifen and raloxifen, promethazine and vitamin K. The author emphasizes the comprehensive character of therapy and the urgent necessity to initiate it early.
