Efficacy of minocycline as a root conditioner in comparison to citric acid and tetracycline. An in vitro evaluation.

This study compared the root surface characteristics produced by Minocycline HCl, Citric acid and Tetracycline HCl. when used as a root conditioner. This study included 5 groups of 10 extracted teeth each with advanced periodontal disease. Diseased root surfaces of group B,C,D and E were planed with Gracy curets and solutions of Citric acid, Tetracycline HCl, Monocycline Hcl and Tween 80 of PH 2.5, were applied to the surface in respective groups with cotton pellets for 5 mts. Group A was not subjected to root planning or application of any root conditioner and was used as a control. The teeth in all five groups were then washed, sectioned processed and examined under scanning electron microscope. These results revealed that the surfaces of acid treated sites differed from each other and with specimens treated sites differed from each other and with specimens treated with root panning alone. The efficacy of Minocycline and tetracycline in the removal of smear layer and exposure of dentinal tubules were comparable and Minocycline HCl though not as effective as Citric acid can probably be recommended in view of its antibactrial activity, anti collagense and substantivity properties.

The authors study in vitro effects of minocycline and compare its activity with tetracycline. 909 strains of cocci and bacilli selected among strains isolated from Clermont-Ferrand Hospital Center during the third trimester 1974 were used. Study involves 3 parts : comparison between bacteriostatic activity of both antibiotics ; evaluation of R-factor resistance ; evaluation of bactericidal activity of antibiotic associations including minocycline. M.I.C. study included all strains. A better bacteriostatic activity was noted with minocycline against multi-resistant strains of staphylococcus, streptococcus, E. coli, Serratia and Moraxella. R-factor resistance has been studied upon 27 strains of Gram-negative bacilli selected for their high M.I.C. to tetracycline (M.I.C. greater than or equal to 64 mcg/ml) and to minocycline (M.I.C. greater than or equal to 32 mcg/ml). Both types of transferable tetracycline resistance characters Ta and Tb have been found. More than 250 bactericidal associations were studied upon 38 strains.

Use of normal C57BL/6 mice with established Mycobacterium avium infections as an alternative model for evaluation of antibiotic activity.

Several murine models have been used to evaluate the activities of antimicrobial agents against Mycobacterium avium infection. The main model used is the beige mouse model, but beige mice are expensive and not easily available. Thus, we developed a model of infection in wild C57BL/6 mice. The drugs that exhibited some activity in a previous model of early infection were evaluated in a new model of established infection. Sparfloxacin (50 mg/kg of body weight), ethambutol (50 mg/kg), minocycline (25 mg/kg), and the inhibitor of the cortisol receptors RU-40555 (100 mg/kg) were compared with clarithromycin (50 mg/kg). Treatments were started 5 weeks after the inoculation and were continued for 21 days. Sparfloxacin and RU-40555, which exhibited a moderate activity in the model of early infection, were not effective in this model of established infection. Clarithromycin and combinations with clarithromycin kept their activities against M. avium infection, both in the spleen and in lungs. The present model of established infection of normal C57BL/6 mice is more relevant than the model of early infection for a stringent evaluation of drugs.
