Sodium depletion and maternal separation in the suckling rat increase its salt intake when adult.

To establish whether neonatal sodium depletion increases the adult's avidity for NaCl, 12-day-old suckling pups were injected with the natriuretic-diuretic furosemide (1 mg) while with their dams. The injections surged plasma aldosterone, and when the rats were adult (70 days), their spontaneous intake of 3% NaCl was increased. Additional experiments investigated whether maternal separation has a similar effect and could thus be a source of individual variation in salt intake of the adult. Fifteen-day-old pups were separated from their dams for 24 h in an incubator. When adult, their intake of 3% NaCl was increased. Availability of saline during maternal separation obviated the effect. The increase in adult intake of 3% NaCl was specific insofar as drinking of water was not increased similarly. The results show that the adult rat's avidity for sodium can be increased by postnatal natriuresis and possibly stress. The implications of the findings are discussed.

Influence of biological matrix on retention behaviour and identification possibilities of selected neutral and acidic drugs in thin layer chromatography. An interlaboratory investigation.

Samples of autopsy blood and liver were spiked individually with aminophenazone, p-aminosalicylic acid, clordiazepoxide, clonazepam, cyclobarbital, furosemide, medazepam, phenacetin, phenazone, and phenobarbital and extracted with diethyl ether at pH 5. The extracts, as well as solutions of pure drugs, were developed in three thin layer chromatographic systems: chloroform:acetone (80:20), ethyl acetate:methanol:ammonia (85:10:5) and chloroform:methanol (90:10). The investigations performed in parallel in two laboratories showed that the intra- and inter-laboratory variability of RF values is larger for drugs extracted from liver. The biological matrix affected both precision and accuracy of results. The number of analysts involved in TLC procedures also affected the intra-laboratory precision.

Effects of angiotensin-converting enzyme inhibition on blood pressure and plasma renin activity in essential hypertension.

Oral converting enzyme inhibitor SQ14225 was administered in 11 patients with essential hypertension, in order to investigate the role of the renin-angiotensin system in the regulation of blood pressure in essential hypertension. In the sodium-repleted state (150 mEq sodium intake for 6 days) in 11 patients, converting enzyme inhibitor decreased the average mean blood pressure from 113 +/- 2 to 106 +/- 2 mm Hg (p less than 0.001). Plasma renin activity increased with sodium depletion (30 mEq sodium intake for 3 days after furosemide treatment) from 1.26 +/- 0.07 to 3.26 +/- 0.48 ng/ml/hr (p less than 0.001). In the sodium-depleted state the hypotensive effect of SQ 14225 was more pronounced (mean blood pressure 108 +/- 2 to 93 +/- 3 mm Hg). The decrease in mean blood pressure caused by the inhibitor correlated to the basal plasma renin activity (r = -0.53, p less than 0.02, n = 22 measurements). The results indicate that the renin-angiotensin system participates in the regulation of blood pressure in essential hypertension, even in the sodium-repleted state. This role of the renin-angiotensin system in blood pressure regulation becomes more crucial during sodium depletion.
