Involvement of amygdalar alpha adrenoceptors in hypothalamo-pituitary-adrenocortical responses.

We have previously demonstrated that the facilitatory effect of the amygdala on the hypothalamo-pituitary-adrenocortical (HPA) axis responses to photic stimulation depends on the presence of norepinephrine (NE) in this region. In the present study we investigated the effect of alpha 1- and beta-adrenoceptor blockers, injected into the central amygdala nucleus, on the HPA responses. In intact freely moving rats, short photic stimulation caused depletion of CRH-41 from the median eminence, due to its release into the portal circulation and a rise in serum ACTH and corticosterone levels. This effect was markedly inhibited by injection of prazosin, an alpha 1-adrenoceptor blocker, but atenolol, a beta 1 blocker, had no effect. These data further indicate that amygdala NE plays a role in the activation of the HPA axis following neural stimuli and that this effect is mediated by alpha 1-adrenoceptors in the central amygdala.

The receptors responsible for heat production in brown adipose tissue in the young rabbit.

It is known that adrenergic agonists stimulate thermogenesis in the brown fat of the young rabbit but the receptors responsible for mediating the response have not been identified. The infusion of either noradrenaline or isoproterenol (1-2 micrograms . kg-1 X min-1) produced an increase in subcutaneous temperature (0.93 +/- 0.15 and 1.22 +/- 0.10 degrees C, respectively over the interscapular brown fat. At low doses (0.4 microgram . kg-1 X min-1) only isoproterenol was effective. The thermogenic response to isoproterenol was blocked by atenolol, a beta 1-adrenergic antagonist. Neither salbutamol or terbutaline, both beta 2-agonists, produced a temperature increase. Collectively, these data suggest that stimulation of beta 1-adrenoceptor is primarily responsible for the thermogenic activity of brown fat in the rabbit. However, it was found that 53% of the increase in temperature could be blocked by prazosin, an alpha 1-antagonist. Phentolamine was not effective as a blocker. Although a maximal brown fat thermogenic response can be achieved by stimulating the beta-adrenoceptors, the alpha-adrenoceptors appears to play at least an auxiliary role in young rabbit.

Analysis of oxprenolol in formulations by high-performance liquid chromatography.

A simple, accurate, and rapid high-performance liquid chromatographic method for the analysis of oxprenolol in commercial formulations is described. The analysis was performed on a cyano radial compression cartridge, with 0.0539 M, pH 3 phosphate buffer-acetonitrile-methanol (76: 15.6:8.4) as the mobile phase. The flow rate was 5 mL/min, with detection at 272 nm; the mobile phase was employed for extraction. The assay was applied to the content uniformity test of three oxprenolol hydrochloride tablet formulations of different strengths and the contents of a 2-mg dry ampule for intravenous/intramuscular injection. The percent of label claim for each formulation tested was within 91.7-110%. The applicability of this assay to the analysis of some other beta-blocking drugs was investigated. It was found that under the above conditions, atenolol, metoprolol, oxprenolol, and propranolol can be fully resolved in less than 3 min.
