Tissue atenolol levels following chronic beta-adrenoceptor blockade using oral atenolol in dogs.

Tissue atenolol concentrations are high following chronic continuous beta-adrenoceptor blockade in dogs. Furthermore, significant concentrations of this poorly lipid soluble drug are found within the central nervous system after chronic dosing. It is suggested that all beta-adrenoceptor blocking agents may enter the central nervous system in significant and sufficient quantities to account for a central antihypertensive action of this group of compounds. Sequestration of beta-adrenoceptor agents in the CNS or other tissues may account for other clinically observed effects including adaptive effects.

Absorption of polar drugs following caecal instillation in healthy volunteers.

We have investigated colonic drug absorption in man by the caecal instillation of a multi-component solution of atenolol, cimetidine, frusemide, hydrochlorothiazide and salicylic acid. We found that salicylic acid absorption from this solution was delayed but complete whereas the absorption of atenolol, cimetidine, frusemide and hydrochlorothiazide was four- to five-fold lower than expected from oral bioavailability studies.

Atenolol in hypertension: follow-up of patients crossed over to this agent from propranolol, pindolol or metoprolol.

Thirty hypertensive patients treated with a drug regimen which contained a beta-receptor antagonist (either propranolol, pindolol or metoprolol) exhibited an improvement in blood pressure control when atenolol was substituted for the corresponding beta-blocker in the drug regimen. Additional benefits noted were a reduction in concomitant drug therapy and an improvement in reported side effects. Atenolol caused a significant decrease in heart rate in the group for which the mean follow-up period was 23.6 months.
