Reversed-phase chromatography is the most common means of separation for small drug molecules. However, polar drugs may suffer from poor retention and peak shape in reversed-phase high-performance liquid chromatography (RP-HPLC). Hydrophilic interaction liquid chromatography (HILIC) provides a viable alternative to RP-HPLC and is an excellent way to separate polar compounds. This paper describes a HILIC/ESI-MS/MS assay for the determination of acyclovir from rat plasma, amniotic fluid, placental tissue, and fetal tissue. The isocratic separation utilizes an underivatized silica column with an acetonitrile/formate buffer mobile phase (80:20). The method is validated over a range of 50 ng/mL to 50 micro g/mL with % error and % relative standard deviation of <15% over 3 days. All samples are prepared by acetonitrile protein precipitation, which yields high recovery (>84% for acyclovir). This assay can be applied to the pharmacokinetic study of the placental transfer of acyclovir. Copyright 2002 John Wiley & Sons, Ltd.

[The effect of Kagocel on herpes virus reproduction]

The effect of an original drug Kagocel on the reproduction of Herpes simplex virus including its mutant strains resistant to basic antiherpetic medicine Acyclovir was evaluated. Kagocel was shown to have a low cytotoxic effect on Vero cell and inhibited reproduction of Herpes virus type 1 and Herpes virus type 2 in noncytotoxic concentrations. Kagocel was also demonstrated to inhibit the reproduction of Herpes virus type 1, resistant to combination of Acyclovir and phosphonoacetic acid. Detection of direct antiherpetic activity of Kagocel in vitro opens good perspectives to its clinical application, especially in combination with Acyclovir, the medication with other antiherpetic mechanism of action.

PIP: AIDS is no myth. It is preventable but there is no cure. But how much are we doing in Africa about those sexually transmitted diseases which are both preventable and curable? I feed sad, really sad, about the cases of men and women who engage in unprotected (without condoms) extra-marital sex and then wait for 2 to 3 weeks before they have sex with their spouses in the belief that, if they have no problems in the meantime, they must be free of infection. They are unaware that it is possible to harbor Neisseria gonorrhoea and other STD organisms in the genital system without having symptoms. Studies have shown that 10% of men and 60% of women infected with N. gonorrhoea have no symptoms. Some who resume a sexual relationship with their spouse after the 2 to 3 week 'watchful period' pass on an infection and then blame their innocent spouse for the consequences] We should seek to improve the knowledge of the public and to disabuse their faith in the watchful period. Another area where action is required concerns herpes genitals. Many doctors are not familiar with atypical presentations of this infection and, consequently, many cases are missed. In view of the importance of early detection and early commencement of therapy with acyclovir (Zovirax), I call upon Wellcome--as manufacturers of Zovirax--to publish and distribute in Africa a chart illustrating the various presentations of the infection. This would help prevent doctors from making such patients go from antibiotic to antibiotic for many months without cure. (full text).
