A novel missense mutation affecting the human hairless thyroid receptor interacting domain 2 causes congenital atrichia.

Congenital atrichias represent a large and heterogeneous group of inherited hair disorders. In this report, we describe a patient affected with alopecia universalis congenita (MIM 203655). Sequence analysis revealed a G to A transition at cDNA position 3034 of the hairless hr gene present in a homozygous state in the patient and in a heterozygous state in the patient's mother, and absent in the patient's sister. The mutation is predicted to result in the substitution of an asparagine residue for an aspartate amino acid (D1012N) at a position previously shown in the rat to affect hairless binding to thyroid hormone receptor. This study presents the first evidence in humans for the functional importance of the hairless thyroid receptor interacting domain 2.

A cross-sectional study of Leishmania spp. infection in clinically healthy dogs with polymerase chain reaction and serology in Greece.

A total of 73 clinically healthy hunting dogs, experiencing an outdoor lifestyle and originating from an area where canine leishmaniasis is endemic, were included in the study. Polymerase chain reaction (PCR) and indirect immunofluorescent antibody test (IFAT) for Leishmania spp. were done on bone marrow and serum samples, respectively, obtained from all 73 dogs, just before the beginning of the sandfly season. PCR was found positive in 46/73 (63%) whereas, IFAT only in 9/73 (12.3%) of the dogs. The prevalence and the incidence of Leishmania infection by PCR were 61.9 and 47.1%, respectively. No association was found between the breed, age, sex, length of hair coat of the dog, urban or rural life and the presence of ample vegetation and water collections in the proximity of their living quarters, and the result of PCR. These findings clearly demonstrate that most of the dogs residing areas where leishmaniasis is endemic become infected but usually remain seronegative. Serological screening of the general canine population in these areas may result in an underestimation of the true prevalence of the infection rate.

Field-amplified sample stacking capillary zone electrophoresis applied to the analysis of opiate drugs in hair.

The present paper describes the methodological optimization and validation of capillary zone electrophoresis (CZE) for the determination of major opiates (morphine, codeine, 6-monoacetylmorphine, acetylcodeine, heroin) in hair samples by using a field-amplified sample stacking injection before the separation in a binary running buffer (0.1 M sodium phosphate, pH 2.5, with 40% ethylene glycol). The applied potential was 20 kV, at 25 degrees C. Detection was by UV absorption at the fixed wavelength of 214 nm or by recording the full spectrum between 190-400 nm, thus improving the analytical selectivity and identification power of CZE. Hair samples were liquid/liquid extracted; dried extracts, reconstituted with a low-conductivity solvent (0.1 mM phosphoric acid, with 80% 1-propanol), were injected by electromigration at 10 kV for 99 s, after a 0.5 mm plug of water. Under the described conditions, the limit of detection (with a signal-to-noise ratio of 3) in hair extracts was 100 pg/mL for codeine, 75 pg/mL for morphine and 6-monoacetylmorphine (6-MAM), 150 pg/mL for ethylmorphine, and 0.75 ng/mL for acetylcodeine and heroin. The precision of the method was validated for standards in pure solution by using internal standardization, providing for intraday and day-to-day assays, in terms of migration times, relative standard deviation (RSD) values < or = 0.2%, and in terms of peak areas, RSD values <5.71%.

Physical and pubertal development in young male gymnasts.

The purpose of this study was to evaluate the effect of intense training on physical growth and sexual maturation in young male gymnasts. Physical development, pubertal development, testosterone levels, energy expenditure, and relative body fat were examined in 21 circumpubertal male gymnasts (13.3 +/- 0.3 yr) and 24 age-matched controls (13.5 +/- 0.3 yr). Subjects completed a self-assessment of genital and pubic hair development with the use of the Tanner scale. All subjects were measured for height, weight, and salivary testosterone levels (T). The Physical Activity Questionnaire for Adolescents was used to estimate weekly energy expenditure in metabolic equivalents. Percent body fat (%BF) was assessed by using bioelectrical impedance analysis. Developmental stages and T, as well as height and weight, were not different between groups. Energy expenditure was significantly higher (P <or= 0.05) and %BF was lower (P <or= 0.05) in athletes than in controls, but lean body mass was not significantly different between groups. Energy expenditure was negatively correlated (P <or= 0.05) with %BF but not related to T. Developmental stages were strongly (P <or= 0.05) related to T but not to energy expenditure or %BF. It is concluded that, although there is a higher energy expenditure accompanying intense training in young male athletes, their body composition is not necessarily affected, and there is no determined effect on their physical and pubertal development.
