Scopulariopsis brevicaulis: a keratinophilic or a keratinolytic fungus?

The morphologic expression of human hair and nail invasion in vitro by Scopulariopsis brevicaulis isolates was studied by light microscopy on whole material and on semi-thin sections, and also by scanning and transmission electron microscopy. Only three isolates of the nine that were examined were keratinolytic, capable of both attacking keratinic substrates and demolishing their keratin. The process showed all the characteristic of enzymatic digestion and was in agreement with the biochemical and morphogenetic scheme proposed for other keratinolytic fungi during their invasion of human hair in vitro. All the active isolates were capable of developing structures related to surface erosion and radial penetration contemporaneously. However the extent and rate of keratinolysis were rather low when compared with the efficiency of other keratinolytic fungi. This finding suggests that S. brevicaulis is of secondary importance in the mineralization of keratinic substrates in natural environments. From the medical standpoint the mere demonstration of keratinolytic activity means that it may be regarded as a real cause of primary infection.

35S-Sulfide incorporation during alkaline treatment of keratin and its relation to lanthionine formation.

Cattle hair exposed to solutions of 35S-sulfide ions at pH 12.5, hydrolyzed with acid, and analyzed for amino acids with simultaneous measurement of the radioactivity of the eluate from the analytical column showed 87 percent of the radioactivity incorporated in the hair in three amino acids: cysteic acid, lanthionine, and cystine. This and other evidence presented lend further support to the intermediacy of dehydroalanyl residues formed by a beta-elimination reaction in the conversion of cystinyl residues to lanthionyl residues in proteins. The presence of radioactively labelled cystine in the hydrolyzate indicates that the dehydroalanyl residues are also capable of reforming cystinyl residues under these same conditions through a series of reversible reactions.

Unusual presentation of Microsporum canis in human hair.

This study reports an unusual presentation of Microsporum canis in infected human hair in a three-year-old boy. Infected hairs showed ectothrix colonization with a mass of small conidia and numerous macroconidia typical of the genus Microsporum. The infected hairs also showed strong invasion by hyphae bearing small numbers of arthroconidia. These hyphae were thinner and longer than those normally found in hair and scalp. It is well known that in cases of tinea capitis due to M. canis, ectothrix arthroconidia formation is extrapilary and arises from intrapilary hyphae. In this case it seems that unusual quantities of hyphae were formed, and not all of them broke down into arthroconidia. They appeared to remain in the hair and to produce macroconidia, which were not formed outside the hair in contact with air, but instead seemed to remain in a deeper layer of the hair than the arthroconidia seen.

Human toxocariasis and direct contact with dogs.

Toxocariasis in man is traditionally thought to be contracted through the ingestion of eggs from contaminated soil. The disease may manifest itself in different syndromes such as ocular larval migrans, visceral larval migrans and covert toxocariasis. This paper assesses the evidence for the soil contamination hypothesis and proposes that direct contact with dogs may provide a better explanation of the epidemiology of the disease. Hair was collected from 60 dogs from various places in Ireland and the UK and examined for the presence of Toxocara canis eggs. T. canis eggs were found in the hair of 25 per cent of the dogs; in total, 71 eggs were recovered, of which 4.2 per cent were embryonated and 23.9 per cent were embryonating. The maximum densities of the embryonating and embryonated eggs were 180 and 20 eggs per gram of hair, respectively, much higher than the densities reported for soil samples. It is suggested that dogs infected with T. canis may infect people by direct contact.

Like developmental biology of any part of our body, hair growth is a complicated process. Hence the homework for modern science to yet unravel the process and mechanism to a completion.
