The transduction of auditory signals by cochlear hair cells depends upon the integrity of hair cell stereociliary bundles. Stereocilia contain a central core of actin filaments, cross-linked by actin bundling proteins. In the cochlea, the two proteins described to date as responsible for the spatial arrangement of actin filaments in sterocilia are fimbrin and the recently discovered espin. Fimbrin (the chick homolog of human I-plastin) belongs to the plastins/fimbrin family that includes two additional isoforms of plastins, T- and L-plastin. In the present study, we used isoform specific antibodies to investigate the presence of the T- and L-isoforms of plastin/fimbrin in the adult and developing rat cochlea. We found that T-plastin, but not L-plastin, is expressed in the rat cochlea. During postnatal development of the rat organ of Corti, T-plastin can be detected in the core of stereocilia from early stages of hair cell differentiation, and its expression gradually increases in stereocilia as hair cells mature. However, as opposed to other actin-binding proteins expressed in stereocilia, T-plastin is absent from the stereocilia of mature hair cells. Such temporally restricted expression strengthens the idea of functional differences between plastins isoforms, and suggests that T-plastin could have a specific role in stereocilia formation. 2002 Wiley-Liss, Inc.

Role of a positive regulator of root hair development, CAPRICE, in Arabidopsis root epidermal cell differentiation.

In Arabidopsis, root hairs are formed only from a set of epidermal cells named trichoblasts or hair-forming cells. Previous studies showed CAPRICE (CPC) promotes differentiation of hair-forming cells by controlling a negative regulator, GLABRA2 (GL2), which is preferentially expressed in hairless cells. Here, we show that CPC is also predominantly expressed in the hairless cells, but not in the neighboring hair-forming cells, and that CPC protein moves to the hair-forming cells and represses the GL2 expression. We also show that the N terminus of bHLH protein interacts with CPC and is responsible for the GL2 expression. We propose a model in which CPC plays a key role in the fate-determination of hair-forming cells.

Environmental conditions in the Rudnaya River watershed--a compilation of Soviet and post-Soviet era sampling around a lead smelter in the Russian Far East.

The Rudnaya River valley in the Russian Far East contains a rich reserve of lead, zinc and boron and has been mined for nearly 100 years. Environmental contamination related to the area's mines and lead smelter was studied for over 30 years during the Soviet era, by members of the Pacific Geographic Institute (PGI). Due to government restrictions, much of the sampling focused on contamination of the river, the air, forests, vegetation, agricultural products and soil. Source-specific samples, such as stack emissions from the smelter, and blood lead levels from the residents and smelter workers could not be obtained or were classified as State secrets. However, the data do describe the extent and severity of the environmental contamination and related public health concerns. Water discharged from the smelter averages 2900 m(3)/day (containing 100 kg of lead (Pb) and 20 kg of arsenic (As)) and leachate from area mine dumps and other industrial processes contaminates the Rudnaya River. Annual air emissions include 85 tonnes of particulates (containing 50 tonnes of Pb and 0.5 tonnes of As) and 250000 m(3) of gases high in sulfur dioxide (SO(2)), carbon monoxide (CO) and carbon dioxide (CO(2)). Vegetative stress is severe and much of this area is denuded. Pb and other metals in agricultural products suggest local produce may be dangerous for human consumption, although it is a major food source for the community. Public and occupational health indicators of basophilic stippling, respiratory disease and hair lead levels further suggest the severity of the problem. Although, descriptions of complete methodologies and procedures are often lacking, these data describe how sampling was conducted during the Soviet era and document a site with severe heavy metals contamination, especially lead, and the likelihood of related public health problems. They are relevant today as investigators employ state-of-the-art-sampling techniques and explore cleanup options under a new governmental system and challenging economic times. In the post-Soviet era, a Russian/US team sampled area soils and dusts and confirmed the severity of the environmental problems using commonly employed sampling and analysis techniques. Lead concentrations in residential gardens (476-4310 mg/kg, Gx=1626 mg/kg) and in roadside soils (2020-22900 mg/kg, Gx=4420 mg/kg) exceed USEPA guidance for remediation. Preliminary biokinetic estimates of mean blood levels (average 13-27 microg/dl) suggest pre-school children are at significant risk of lead poisoning from soil/dust ingestion. Today, the PGI, in cooperation with the industrial owners and the local health and environmental authorities, is attempting to establish long-term monitoring and pollution abatement within the constraints of their difficult economic situation.

A simple, but sensitive and specific high-performance liquid chromatographic assay for the simultaneous determination of the major constituents of "ecstasy" [i.e. 3,4-methylenedioxymethamphetamine (MDMA), 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxyethylamphetamine (MDE)] with direct fluorimetric detection, particularly intended for the routine analysis of hair, is described. Hair samples (100 mg) were overnight incubated in 1 ml of 0.25 M HCl at 45 degrees C and extracted with a commercial liquid-liquid method. The dried residue reconstituted with 500 microl of 0.05 M NaH2PO4 pH 5.2 was injected. Isocratic reversed-phase liquid chromatography was carried out on a column (250x4.6 mm I.D.) packed with spherical 5-microm poly(styrene-divinylbenzene) particles; the mobile phase was composed of 0.1 M potassium phosphate (pH 3)-acetonitrile (82:18). The excitation and the emission wavelengths were set to 285 and 320 nm, respectively. Under the described conditions, MDA, MDMA and MDE eluted in symmetric peaks with an analysis time of 30 min. The limit of detection was lower than 1 ng/ml, with a signal-to-noise ratio of 5, for each compound in solution, allowing a cut-off of 0.1 ng/mg in the hair matrix to be established. The intra-day precision (n = 6) of the assay was characterised by RSDs between 1.0 and 3.0% and between 0.52 and 0.88% for concentrations of 10 and 100 ng/ml, respectively; in day-to-day precision tests (n = 6), RSDs ranged between 5.12 and 11.12%, respectively, for the same concentrations. Interferences from as many as 92 therapeutic and/or abused drugs currently in use in the population were excluded, including N-methyl-1-(3,4-methylenedioxyphenyl)-2 butanamine (MBDB).
