One of the major obstacles hindering the progress of studies on mammalian cochlear physiology is the inaccessibility of inner ear cells located in a complex structure of the bony labyrinth. We describe here a technique to record cellular fluorescent signals from any identified inner ear cells in cochlear slices and hemicochleae. Cochlear slices were obtained from postnatal rats (P0-P7) before the cochlea completely ossify, and hemicochleae were cut from older animals (P7-adult). Individual inner ear cells were visually identified using infrared differential interference contrast or oblique illumination optics. Techniques were developed for either bulk-loading cells or loading selected single cells with Ca(2+) indicator dyes, and for maintaining functional viability of cochlear slices/hemicochleae for recordings. Robust and reliable responses of ligand-gated receptors were recorded from individual inner ear cells (e.g. hair cells, spiral ganglion neurons etc.) for at least 24 h after slices/hemicochleae were cut by an oscillating tissue slicer. The technique described here allowed direct observations of [Ca(2+)](i) activities from multiple cells simultaneously in situ, thus providing a feasible way to study the intercellular communication or networking activities from identified cells in the inner ear.

Trichoblastic neoplasm with apocrine differentiation.

Trichoblastoma is a benign neoplasm with primitive hair follicle differentiation. The tumor is characterized by nests and cords of epithelial cells in an organized relation to stroma. The epithelial cells differentiate toward follicular germ and follicular sheath. More divergent differentiation in the form of sebaceous elements in trichoblastoma has infrequently been reported in the literature. We report a trichoblastic neoplasm showing typical features of trichoblastoma as well as areas of apocrine differentiation. The neoplasm, which was present for several months, was removed from the chin of a 31-year-old woman. Histologic examination showed a neoplasm composed of basaloid nests with occasional papillary mesenchymal bodies as well as ducts and glands exhibiting apocrine differentiation. Trichoblastoma with apocrine features is rarely reported in the literature.

Epidermal cells in the root of Arabidopsis seedling differentiate either as hair or non-hair cells, while in the hypocotyl they become either stomatal or elongated cells. WEREWOLF (WER) and GLABRA2 (GL2) are positive regulators of non-hair and elongated cell development. CAPRICE (CPC) is a positive regulator of hair cell development in the root. We show that WER, GL2 and CPC are expressed and active during the stages of embryogenesis when the pattern of cells in the epidermis of the root-hypocotyl axis forms. GL2 is first expressed in the future epidermis in the heart stage embryo and its expression is progressively restricted to those cells that will acquire a non-hair identity in the transition between torpedo and mature stage. The expression of GL2 at the heart stage requires WER function. WER and CPC are transiently expressed throughout the root epidermal layer in the torpedo stage embryo when the cell-specific pattern of GL2 expression is being established in the epidermis. We also show that WER positively regulates CPC transcription and GL2 negatively regulates WER transcription in the mature embryo. We propose that the restriction of GL2 to the future non-hair cells in the root epidermis can be correlated with the activities of WER and CPC during torpedo stage. In the embryonic hypocotyl we show that WER controls GL2 expression. We also provide evidence indicating that CPC may also regulate GL2 expression in the hypocotyl.

The PUVA-turban as a new option of applying a dilute psoralen solution selectively to the scalp of patients with alopecia areata.

BACKGROUND: Alopecia areata is a burden for many patients and often resistant, even to extensive therapy. Orally administered PUVA therapy has been shown among numerous systemic and topical treatment modalities to be a therapeutic alternative. However, the clinical use of oral PUVA is often limited by systemic side effects. Bath-PUVA therapy offers an alternative solution because of the negligible systemic absorption of psoralen with this technique. Through use of a "PUVA-turban" it is now possible to administer a dilute bathwater solution containing 8-methoxypsoralen (8-MOP) to the scalp. OBJECTIVE: The purpose of this study was to determine whether PUVA turban therapy is effective in treating alopecia areata in different clinical stages. METHODS: We treated 9 patients with severe, rapidly progressing, treatment-resistant alopecia areata with PUVA-turban treatment as a modification of bath-PUVA therapy. At each treatment session a cotton towel was soaked with a 0.0001% 8-MOP solution (1 mg/L) at 37 degrees C, wrung gently to remove excess water, and wrapped around the patient's head in a turban fashion for 20 minutes. This was directly followed by UVA radiation. Treatment sessions were initially performed 3 to 4 times per week. RESULTS: The cumulative UVA doses given over treatment periods of up to 24 weeks were 60.9 to 178.2 J/cm(2), with single doses ranging from 0.3 to 8.0 J/cm(2). After up to 10 weeks of treatment, hair regrowth could be noticed in 6 of 9 patients. Two patients did not respond to the treatment, and one patient showed only vellus hair regrowth. CONCLUSION: PUVA-turban therapy can be considered a useful method of administering a dilute psoralen solution selectively to the scalp of patients. It has been shown to be a well-tolerated and, in some patients, efficient therapeutic alternative in the treatment of alopecia areata.
