Twelve non-nodulating pea (Pisum sativum L.) mutants were studied to identify the blocks in nodule tissue development. In nine, the reason for the lack of infection thread (IT) development was studied; this had been characterized previously in the other three mutants. With respect to IT development, mutants in gene sym7 are interrupted at the stage of colonization of the pocket in the curled root hair (Crh- phenotype), mutants in genes sym37 and sym38 are blocked at the stage of IT growth in the root hair cell (Ith- phenotype) and mutants in gene sym34 at the stage of IT growth inside root cortex cells (Itr- phenotype). With respect to nodule tissue development, mutants in genes sym7, sym14 and sym35 were shown to be blocked at the stage of cortical cell divisions (Ccd- phenotype), mutants in gene sym34 are halted at the stage of nodule primordium (NP) development (Npd- phenotype) and mutants in genes sym37 and sym38 are arrested at the stage of nodule meristem development (Nmd- phenotype). Thus, the sequential functioning of the genes Sym37, Sym38 and the gene Sym34 apparently differs in the infection process and during nodule tissue development. Based on these data, a scheme is suggested for the sequential functioning of early pea symbiotic genes in the two developmental processes: infection and nodule tissue formation.

Cats require more dietary phenylalanine or tyrosine for melanin deposition in hair than for maximal growth.

In 1986, the NRC recommended a dietary concentration of 4.0 g/kg of phenylalanine and 8.5 g/kg of total aromatic amino acids for growing kittens on the basis of maximal growth rate and nitrogen balance. Black hair-coated cats were given purified diets containing the following phenylalanine + tyrosine (Phe + Tyr) concentrations (g/kg diet): 4 + 2, 4 + 4, 4 + 6, 4 + 8, 10 + 0, 10 + 2, 10 + 4, 10 + 6, 10 + 8, 10 + 10, 24 + 0 for at least 6 mo. All other amino acids were present at about twice the requirements. Total melanin and the oxidation product of eumelanin, pyrrole-2,3,5-tricarboxylic acid (PTCA) were measured in hair. There was a significant linear relationship between the concentrations of tyrosine in plasma and PTCA in hair. The relationship between PTCA concentration in hair and Phe + Tyr in the diet had a point of inflection at approximately 16 g/kg Phe + Tyr. Cats fed diets with <16 g Phe + Tyr developed "red hair." We confirmed the anecdotal reports that the black hair of cats can change from black to reddish brown. An aromatic amino acid concentration > or =18 g/kg is recommended for the prevention of visually discernible red hair in black-coated cats. Dietary concentrations >18 g total aromatic amino acids/kg diet promote a greater ratio of PTCA:total melanin in hair. We are unaware of a secondary nutrient requirement being so much greater than the requirement for growth.

Influence of tyrosinase levels on pigment accumulation in the retinal pigment epithelium and on the uncrossed retinal projection.

To study the relationship among tyrosinase activity, melanin production, and the routing of retinal ganglion cell (RGC) axons at the optic chiasm, we analysed mice with varying doses of the tyrosinase gene. These include the dark-eyed albino (Tyrc44H), a radiation-induced hypomorphic allele of tyrosinase; and transgenic mice carrying 1 or 2 alleles of a tyrosinase minigene on both wild-type (Tyr+) and albino (Tyrc) backgrounds. Melanization of the retinal pigment epithelium (RPE) occurred gradually even at <2% wild-type tyrosinase activity and was sensitive to tyrosinase activity up to <35% of wild-type levels, beyond which melanin synthesis appeared to be saturated. Overexpression of tyrosinase led to tyrosinase activity above wild type level, but did not increase melanin production. Although a loss of melanin because of a mutation in tyrosinase is associated with a decrease in the number of uncrossed fibers, elevating tyrosinase levels does not appear to cause an increase in the size of the uncrossed retinal projection. Our results suggest that replacing less than 35% of wild-type tyrosinase activity is sufficient to restore normal pigmentation of the RPE, and potentially, to allay visual defects.
