In plants, morphogenesis is largely determined by the orientation and extent of cell enlargement. To define the molecular mechanisms regulating plant cell enlargement, we have conducted a molecular genetic analysis of the ROOT HAIR DEFECTIVE3 (RHD3) gene of Arabidopsis thaliana. Mutations affecting the RHD3 gene were found to alter cell size, but not cell number, in tissues throughout the plant. Genetic and physiological analyses suggest that the RHD3 gene is not required for proper cell type specification, and it is likely to act downstream of the hormones auxin and ethylene. The RHD3 gene was cloned by a T-DNA tagging method and confirmed by the molecular complementation of the rhd3 mutant phenotype and by the analyses of six rhd3 mutant alleles. Consistent with the global effects of the rhd3 mutations, the RHD3 gene is expressed in all major plant organs. The deduced RHD3 product is a novel 89-kD polypeptide with putative GTP-binding motifs near the amino terminus. RHD3-like genes were identified from a protozoan (Entamoeba histolytica), a fungus (Saccharomyces cerevisiae), and another plant species (Oryza sativa), with the sequence identity including the putative GTP-binding motifs. These results imply that the RHD3 protein is a member of a new class of GTP-binding proteins that is widespread in eukaryotes and required for regulated cell enlargement.

Ontogenesis of rat cochlea. A quantitative study of the organ of Corti.

A systematic quantitative set of data concerning the organ of Corti in developing Sprague-Dawley rats at intervals from 18 days of gestation to 10 days after birth (DAB) is provided in this study. Using phalloidin staining, the total number of inner and outer hair cells, the whole length of cochlea, as well as the diameter of inner and outer hair cells and the intercellular space between inner hair cells were determined in order to analyze the quantitative change of inner and outer hair cells during development and to explore some roles of the factors regulating the growth of cochlea. The results show that: (1) The length of cochlea approached its adult size by 7DAB. (2) The growth of the extreme part of the apex was responsible for the delayed elongation of the cochlea. (3) Growth in the cochlear length mainly results from an increase of cell diameter tempered by a decrease of intercellular space. (4) The adult size of inner and outer hair cells was obtained by 7-14DAB. (5) The final number of inner and outer hair cells was reached at 3DAB and remained constant through adulthood. No significant hair cell overproduction and cell death were observed during ontogenesis of the cochlea. The negligible importance of overproduction and missing hair cells during hair cell differentiation suggest that there is a precise regulation phenomenon for producing the right spatial organization of the organ of Corti.

Rab3A immunolocalization in the mammalian vestibular end-organs during development and comparison with synaptophysin expression.

Rab proteins are essential for membrane vesicle docking and fusion and for transport vesicle formation at the presynaptic membrane, a step in the release of neurotransmitters. The vestibular sensory epithelia contain three types of synapses: afferent terminals, efferent endings and possible synaptic contacts between the apex of the afferent nerve calyces and the sensory cells. We report an immunocytochemical codetection of rab3A and synaptophysin in the vestibular end-organs of mouse, between fetal day 14 and adult, and of rat during the postnatal development. During mouse fetal development, rab3A appeared in afferent neurites on F16, and in sensory cells on F19. This was respectively two and five days later than the appearance of synaptophysin-IR in the same compartments. During the late postnatal development and in the adult sensory epithelia, rab3A and synaptophysin were strongly detected in nerve terminals of efferent and possibly afferent nature and in the upper part of the nerve calyces. The presence of rab3A in the nerve calyces is consistent with the putative secretory function of the calyx. In addition, rab3A immunostaining was also present in the sensory cells together with a faint synaptophysin-IR, that had not been described in previous reports [Scarfone, E., Dememes, D. and Sans, A. J. Neurosci., 11 (1991) 1173-1181.]. The presence of these two proteins in the sensory cells supports the existence of a synaptic vesicle cycle in these cells.
